En la NGS, donde el objetivo final es generar bibliotecas muy complejas y secuenciarlas con la m\u00e1xima profundidad y homogeneidad, la cuantificaci\u00f3n precisa es especialmente importante. Los dos pasos clave para determinar con precisi\u00f3n las concentraciones de \u00e1cidos nucleicos son:<\/p><ol><li>La eficacia m\u00e1xima de la enzima es sensible a la cantidad de ADN introducido antes de la preparaci\u00f3n de la biblioteca y 2) despu\u00e9s de la preparaci\u00f3n de la biblioteca para estandarizar las muestras y agruparlas antes de la secuenciaci\u00f3n. Si no se mezclan bien, las lecturas variar\u00e1n de un pocillo a otro. Evite tambi\u00e9n la introducci\u00f3n de burbujas de aire, que pueden interferir con la fluorescencia.<\/p><\/li><li><p>Incubar el ensayo a temperatura ambiente durante 5 minutos.<\/p><\/li><li><p>Detecci\u00f3n de fluorescencia (504nmEx\/531nmEm).
